Luke played with Bohemians' A Championship side in 2011 and graduated to the first team in 2012 under new manager Aaron Callaghan. He made his senior debut for Bohs in a Setanta Cup tie against Portadown on 13 February 2012 and went on to make 17 appearances for Bohemians in the league as the club finished seventh in the Premier Division. He also played both European games against Icelandic team Thor Akureyri, which Bohs lost 5-1 on aggregate and was selected for the Ireland U19 squad during the year.

Luke re-signed with the club for the 2013 season and became a key member of the Bohemians team, playing in 31 games during the league season as the Gypsies finished tenth in the table, just two points away from the relegation play-offs.

In November 2013 Luke crossed the Liffey to sign for Trevor Croly at Shamrock Rovers. In his first season at the club, Luke made 22 league appearances at left-back as Rovers secured fourth place in the 2014 Premier Division table. Under Pat Fenlon in 2015 he made another 21 league appearances, scoring once in a 2-1 win over Galway United, as Shamrock Rovers finished third in the Premier Division. He also played in all four of the club's Europa League matches as Rovers went out to Norwegian side Odds.

In 2016, Luke's third season, at Shamrock Rovers he had made just 1 league appearance when a Cruciate ligament injury brought an abrupt end to his season. Luke re-signed for the Hoops for the 2018 season. In October 2018, Luke announced he was to leave the Hoops after five years at the club.
